Histochemical study of innervation and NADPH-D activity of the thymus.

Acetylcholinesterase (ACHE)-positive nerve fibres in the thymus of the rat form perivascular plexuses as well as free ACHE-positive nerves, which do not follow the course of the vessels. A higher number of ACHE-positive nerves was observed in corticomedullary border and the medulla, a lower number in the cortex of the thymus lobuli. A gradual reduction of ACHE-positive intrathymus nerve formation was recorded and correlated with the increasing age of the rats. Nicotinamide adenine dinucleotide phosphate-diaphorase positive cells observed in the rat thymus were irregular with numerous projections and were located on the boundary of the cortex and medulla.

Manipulations of ACHE gene expression suggest non-catalytic involvement of acetylcholinesterase in the functioning of mammalian photoreceptors but not in retinal degeneration.

To explore role(s) of acetylcholinesterase (AChE) in functioning and diseased photoreceptors, we studied normal (rd/ ) and degenerating (rd/rd) murine retinas. All retinal neurons, expressed AChEmRNA throughout fetal development. AChE and c-Fos mRNAs peaked at post-natal days 10-12, when apoptosis of rd/rd photoreceptors begins. Moreover, c-Fos and AChEmRNA were co-overexpressed in rd/rd mice producing transgenic human (h), and host (m) AChE, but not in rd/ mice. However, mAChE overexpression also occurred in transgenics expressing human serum albumin. Drastic variations in AChE catalytic activity were ineffective during development. Neither transgenic excess nor diisopropylfluorophosphonate (DFP) inhibition (80%) affected the rd phenotype; nor did DFP exposure induce photoreceptor degeneration or affect other key cholinergic proteins in rd/ mice, unlike reports of adult mice and despite massive induction under DFP of c-Fos70 years). Therefore, the extreme retinal sensitivity to AChE modulation may reflect non-catalytic function(s) of AChE in adult photoreceptors. These findings exclude AChE as causing the rd phenotype, suggest that its primary function(s) in mammalian retinal development are non-catalytic ones and indicate special role(s) for the AChE protein in adult photoreceptors.

[Acetylcholinesterase-positive innervation in the regenerated thymus in old rats after surgical and chemical castration].

Surgical and chemical castrations in old rats result in the regeneration of the involuted thymus. The distribution of ACHE-positive nerves did not differ in the regenerated thymi after surgical and chemical castrations. A number of ACHE-positive nerves were detected not only in perivascular topography but also in the functioning thymus parenchyma. A marked difference in thymus density of ACHE-positive nerves was found in rats without castration. We assume that our findings represent an indirect evidence that cholinergic nerves stimulate lymphopoiesis in the thymus. (Fig. 6, Ref. 16.)

[Acetylcholinesterase-positive innervation of the thymus in old rats after ovariectomy].

Surgical ovariectomy in aged rats entails regeneration of the involuted thymus. These results correlate with our study of acetylcholinesterase positive innervation of regenerated thymuses. We found out a marked autonomous innervation of the perivascular tissues, as well as that of the functional parenchyma of the thymus. We proved a significant increase in the density of ACHE-positive nerves in the thymus of aged rats after surgical ovariectomy. These results are considered to represent and indirect evidence of the fact that cholinergic nerves stimulate lymphopoiesis in the thymus. (Fig. 5, Ref. 9.)
